I need help with solving page 122 and 123. I will attach the lab report and gel image. (Note any bands greater than 5Kb are contaminating chromosomal DNA in the sample and do not need to be identified with size, topology and conformation in the homework table.

1.To know the size of the bands of the ladder on your gel image, look at the pattern of the ladder bands in the lab manual and compare to your gel image.

2.First thing- Look at the middle two bands which are 1650bp and 2000bp.

3.Then move up from the middle bands to as far as can be resolved and assign values to those bands.I think you will only be able to resolve up to about 6000bp or 7000bp.

4.Then move down from the middle bands to as far as can be resolved and assign values to those bands.I think you could probably be able to resolve bands down to about 300bpBe sure to attach a graph of your standard curve to your submission. Clearly show the interpolation on the graph or the equation of the trendline if you are calculating size of the unknown bands on your gel.

5.Now that you know the size of each of the bands on your gel image, measure the distance each of those bands in the ladder have traveled from the start of the gel (the well).

6.Now you have distance migrated for each of the bands ON YOUR GEL IMAGE and you know the size of each band by comparing the pattern of the ladder image in your lab manual p92 to your gel image ladder.

7.Now it is time to make the standard curve graph.
